My brother in law's 50th birthday is coming up.

I am wondering whether there is any software that allows a Mac or iPod
to be used like a Jukebox so that people can just use a mouse to select
music by clicking on buttons on a screen.

I could put music into various categories and then write an applescript
to do this (hopefully iTunes is fully scriptable), but if someone has
written a program to do it already, why bother. Anyone know of any
software that has this capability?

You could set up Sound Byte
(http://www.blackcatsystems.com/software/soundbyte.html)
to do the job, but the free lite version only allows for 75 tracks.  You
might need to purchase a license for the regular or pro versions to get
enough songs up for a party.
